  • Expression of mRNA of vascular endothelial growth factor in mouse with oxygeninduced retinopathy

    ObjectiveTo analyze the regulative rule of mRNA of vascular endothelial growth factor (VEGF) in mice with oxygen-induced retinopathy, and to elucidate the possible mechanism of occurrence of neovascularization in retinopathy of prematurity (ROP).MethodsSixty 7-day-old C57BL/6J mice were divided into oxygen-induced retinopathy group and control group. In oxygen-induced retinopathy group, 36 mice were exposed to 75% oxygen for 5 days and then to room air for 5 days; in control group, 24 mice were raised in room air. Vascular perfusion of fluorescein and retinal stretched preparation were used to observe the morphologic changes of retinal vessels. Reversal transcriptionpolymerase chain reaction (RT-PCR) was used to observe changes of VEGF mRNA in each group. ResultsIn oxygen-induced retinopathy group, the morphologic characteristics of retinal vessels were the unperfused area at the center of superficial and deepseated vessels, and the neovascularization appeared at mid-peripheral retina after 2 days in relative hypoxia condition. The results of RT-PCR showed space-time corresponding relation between expression of VEGF and neovascularization, which meant that the transcription of VEGF mRNA decreased in hyperxia conditionand increased in relative hypoxia condition. ConclusionHypoxia is the main reason of occurrence of retinal neovascularization; increased expression of VEGF caused by relative hypoxia after hyperxia might be effective in reducing the occurrence of neovascularization in ROP.(Chin J Ocul Fundus Dis, 2005,21:292-295)

  • Effect of vascular endothelial cell growth factor (VEGF) antisense oligodeoxynucleotides on expression of VEGF in rats with oxygen-induced retinopathy

    Objective To evaluate the effect of vascular endothelial cell growth factor (VEGF) antisense oligodeoxynucleotides (ASODNs) on the expression of VEGF in rats with oxygen-induced retinopathy. Methods Thirty newborn Sprague-Dawley (SD) rats were randomly divided into 3 groups:normal control group, disposal group and non-disposed group, The animal models with oxygen-induced proliferative retinopathy were established by raising the rats in hyperoxic environment. Retrobulbar injection was performed with VEGF ASODNs or normal saline on the rats in 3 groups respectively. The intraocular tissues (all the tissues except the cornea, sclera, and lens) and serum were collected, and the expressions of VEGF were determined by using competitive enzyme immunoassay.Results The expressions of VEGF in intraocular tissues of rats in disposal group were significantly lower than those in non-disposed group (P<0.05), and there was no statistical difference between the disposal and normal control group (P>0.05). There was no significant difference of the expressions of VEGF in serum of rats between the disposal and non-disposed group (P>0.05), which were both lower than those in the normal control group (P<0.05). Conclusion VEGF ASODNs could significantly inhibit the expression of VEGF in intraocular tissues. (Chin J Ocul Fundus Dis,2003,19:172-174)

  • Expression of pax-6 in retina of eyes with myopia induced by optical defocus

    Objectives To investigate the expression of pax-6 in ret ina of in fant monkeys with myopia induced by optical defocus, and to determine the role of pax-6 would play or not in onset and development of myopia and emmetropization.Methods Nine healthy infant rhesus monkeys, aged from 1 to 3 months, were selected and wore spectacle lenses or underwent photorefractive keratectomy (PRK).Transcription polymerase chain reaction method and quantitative analysis were used to determine the expression of pax-6 in the retina with myopia induced by optical defocus in different time, and the result was compared with that in retina without myopia.Results The myopia caused by hyperopic defocus was found. The expression of pax-6 in the retina with myopia induced by optical defocus was significantly higher than that in the retina without myopia(t=3.480,P=0.004).Conclusions The expression of pax-6 is enhanced by hyperopic defocus in the infant monkey retina, which suggests that pax-6 may be involved in vision-dependent eye growth and emmetropization.   • Expression of T cell receptor Vβ8.3 gene in rats with experimental autoimmune uveoretinitis

    Objective To investigate the expression of T cell receptor (TCR) Vβ8.3 gene on CD4+ T lymphocytes in the rats with experimental autoimmune uveoretinitis (EAU). Methods Eighteen Lewis rats were divided into EAU, complete Freund′s adjuvant, and the control group. Inter photoreceptor retinoid-binding protein (IRBP) R16 peptide was synthesized using Fmoc procedure for induction of EAU. Magnetic absorption cell sorting (MACS) me thod was used to isolate the CD4+T lymphocytes from the spleen of the rats. Flow cytometry was used to monitor the efficiency of isolation. The expression of TCR Vβ8.3 gene segment on CD4+T lymphocytes was determined by fluorescent quantitative polymerase chain reaction. Results EAU was successfully induced in the Lewis rats immunized with IRBP R16 peptide. The proportion of CD4+T lymphocytes isolated by means of MACS was statistically higher than that before isolation (P<0.001). The expression of TCR Vβ8.3 gene segment on CD4+ T lymphocytes in EAU rats was significantly higher than that in the control (P<0.05). Conclusions There is a predominant usage of antigen-specific TCR Vβ 8.3 gene in EAU rats induced by IR BP R16 peptide, which may serve as a target for immunotherapy of EAU.   • Interleukin-1α induces retinal neovascularization and the expression of vascular endothelium growth factor in SD rats

    Purpose To determine the effect of exogenous interleukin-1alpha; (IL-1alpha;) on the retina and its vasculature and VEGF expression in SD rats. Methods IL-1alpha;2.0 ng (20 mu;l)  were injected into the vitreous of 8 left eyes of 8 SD rats while steriled PBS were injected into 8 right contralateral eyes of the same rats as control. All eyes were assessed by direct ophthalmoscopy every day and enucleated on the 7 thpostoperative day. Histological examination (hemato xylineosin staining) and immunohistochemical staining with antibody against VEGF antigen were performed, and sections were observed and photographed under light microscopy. Results ①All 8 IL-1alpha; inject ed eyes developed epiretinal membranes and extraretinal neovascularization on the 3 rd postoperative days while none of the 8 control eyes exhibited any a bnormal retinal vascular changes and they were confirmed by HE staining;②Immuno staining identified VEGF express mainly in the inner layer of vessel walls, the epiretinal membranes, the neuroganglional layer and the photoreceptor layer of retina, while the control eyes showed only weak positive staining in the photo receptor layer. Conclusions IL-1alpha; is capable of inducing vitreo retinal neovascularization,and increasing the expression of VEGF in the retina and epiretinal membranes. (Chin J Ocul Fundus Dis, 2001,17:135-137)

  • Induction and maintenance of immune deviation in different intraocular compartment

    Objective To determine whether the vitreous cavity and the subretinal space of different species animals support the induction of immune deviation to soluble antigen and to observe its maintenance time. Methods Bovine serum albumin(BSA)was used as a soluble antigen and inoculated into the anterior chamber(AC),the vitreous cavity(VC)and subretinal space of different animals(rat,rabbit and monkey)respectively.Recipient animals were immunized with BSA and complete Freundrsquo;sadjuvant. Delayed-type hypersensitivity(DTH)was assessed by skin challenge.The maintenance time of deviant immune response was evaluated in the fixed time interval. Results All the animals receiving intraocular antigen inoculation did not display DTH reaction.The maintenance time of immune deviation after inoculation of antigen in different sites is (1)anterior chamber:70 days in rat,90 days in rabbit,320 days in monkey;(2)vitreous cavity:100 days in rat,150 days in rabbit,360 days in monkey;(3)subretinal space:50 days in rat,70 days in rabbit,300 days in monkey. Conclusions The maintenance time of immune deviation varied with different species of animals and different intraocular compartment.The evolution of organisms is synchronous with that of immune system.   • Retinal mRNA expression of NMDAR1 gene in acute elevated IOP models of rabbits

    Objective To quantify the mRNA expression of NMDAR1 gene in the retina of eyes with acute elevation of IOP in rabbit. Methods Tweenty-six eyes of 16 rabbits were divided into three groups: Group 1: The IOP of one eye in 10 rabbits was elevated to 60 mm Hg by ante ri or chamber infusion. Group 2: The another eye of the same rabbit in group 1 was maintained the IOP to 20 mm Hg by anterior chamber infusion. Group 3: Unilat eral eyes of six rabbits were enucleated to evaluate the mRNA levels as normal control group. PCR product was identified by Southern blotting and the mRNA expression level was quantified by RT-PCR. Results The results revealed no significant difference between group 1 and group 2. Conclusion This implies that acute elevated IOP may not affect the mRNA expression level of NMDAR1 gene. (Chin J Ocul Fundus Dis, 2001,17:50-51)

  • Effect of inducible nitric oxide synthase or cyclooxygenase-2 on retinal neovasc ularization in oxygeninduced retinopathy mouse model

    Objective To investigate the effect of inducible nitric oxide synth ase (iNOS) or cyclooxygenase-2 (COX-2) on retinal neovascularization and its possible mechanism in oxygen-induced retinopathy (OIR) mouse model. Methods Retinal neovascularization was induced by oxygen with different concentration. The expression of iNOS, COX-2, matrix metalloproteinases 2 (MMP-2) and vascular end othelial growth factor (VEGF) in the retinae of experimental animals were analyzed by immunohistochemistry, realtime polymerase chain reaction and western blotting technologies. Results The inhibition of COX-2 or iNOS obviously attenuated retinal neovascularization and decreased the expression of VEGF and MMP-2. The iNOS inhibition decreased COX-2 expression, and vice versa. Conclusions COX-2 and iNOS may play a role in retinal neovascularization in OIR mouse model, which may act by regulating the expression of VEGF and MMP-2.

  • Establishment of rabbit models of mixture-infectious endophthalmitis induced by staphylococcus aureus and Escherichia coli

    Objective To establish rabbit models of mixture-infectious endophthalmitis induced by exogenous Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli). Methods A total of 84 eyes of 42 New Zealand white albino rabbits were randomly divided into 4 groups. There were 21 eyes in each group. Rabbit eyes in group 1, 2, 3 and 4 received an intravitreal injection of 0.1 ml of mix bacterium (2times;104 CFU/ ml, including 103 S. aureus and 103 E. coli), S. aureus (104 CFU/ ml), E. coli (104 CFU/ml), and sterilized saline respectively. The eyes were examined by slit-lamp microscopy, ophthalmoscopy, A/B scan, electroretinography (ERG) and bacterial culture of vitreous humors at the timepoints of 6, 12, 24, 48 and 72 hours, and 4, 7, 10, 14 days after intravitreal injection. All eyeballs were then enucleated for histopathological examination. Results Various degrees of inflammatory reactions were presented in the 3 experimental groups after the injection, and the development trend of the disease was nearly the same. In group 1 active intraocular inflammation like anterior chamber exudates, started at 12 hours after injection (which was early than that in group 2 and 3), aggravated between 48 and 72 hours, alleviated slowly from 4 to 7 days, and was obviously better after 10 to 14 days while the corneal neovascularization and vitreous gray opacity begun to form. The bacterial culture was positive in group 1 (100%, 6 hours to 14 days after injection), group 2 (100%, 6 hours to 3 days after injection) and group 3 (100% from 6 hours to 7 days, and 67.67% at 14 days after injection). It was negative for group 2 (7 to 14 days after injection) and group 4 (6 hours to 14 days after injection). The amplitude of ERG b wave dissapeard in group 1 to 3, and decreased less than 30% in group 4 from the 48th hour after injection. Histopathological examination revealed that all intraocular structures infiltrated with inflammatory cells. Conclusion Complicated endophthalmitis rabbit models can be successfully established by intravitreal injection with S. aureus and E. coli.

  • Experimental study of autoimmune uveitis induced by urea soluble fraction of bovine melanin-associated antigen

    Purpose To investigate whether experimental autoimmune uveitis can be induced equally in different rats by urea soluble fraction of bovine melanin-associated antigen(USF-BMAA),and,if so,difference among them. Methods Lewis rats,F344 rats,Wistar rats were immunized with USF-BMAA emulsified with complete Freud is adijuvant and Bordelella pertussis to induce experimental autoimmune uveitis.The animal models were investigated clinically and histopathologically and compared with each other. Rusults Experimental autoimmune uveitis could be induced in Lewis rats,F344 rats and Wistar rats with US-BMAA.Clinical and histopathalogical examination showed that bilateral ocular inflammation developed after immunization 9-13 days.Although inflammation was mainly located in anterior uvea,a mild focal choroiditis was noted in those with severe anterior inflammation.No inflammation was observed in the retina and pineal gland.Experimental autoimmune uveieis induced with USF-BMAA was similar to experimental autoimmune anterior uveitis incited with BMAA presented by other authors.Inflammation induced with USF-BMAA in F344 rats and in Lewis rats was quite similar in the severity and course of the model.But the inflammation was less in Wistar rats compared with that in Lewis rats and F344 rats. Conclusion Experimental autoimmune anterior uveitis was successfully induced with USF-BMAA in Lewis rats,F344 rats and Wistar rats.The difference with regard to the severity among these aminals were propably attributed to their genetic bankground.
