  • EXPERIMENTAL STUDY OF PERIOSTEUM AUTOGRAFT IN REPAIRING BONE DEFECT AND ITS CLINICAL APPLICATION

    Abstract In order to find a new method to repair large bone defect, the free periosteum autograft was investigated in experiment, and then the method was used clinically. In the experiment, a 6mm×18mm×5mm bone defect was made at upper end of both tibiae of 42 rabbites. The periosteum of each rabbit was cut into 1mm cubes, and implanted randomly into the tbial bone defect on one side and the other side was used as control. After 2, 4, 8 weeks, the bone defects of each group were examined for bone formation by roentgenography, radionuclide and histology. The results showed that the defects treated by free periosteum autografts healed twice as fast as the controls (its natural healing). The reason probably was that the periosteum provided with many osteogenic cells. On thebasis of these results, 21 cases of bone defects (the largest was 10.5cm×4cm×4cm, the smallest was 2cm×2cm×2cm) including 17 cases of benign bone tumor and4 cases of chronic osteomyelitis, were treated by free periosteum autografts. The defects were all healed, and the function of the joints was restored.

  • STUDY ON EAU AND EAP MEDIATED BY ADOPTIVE TRANSFER OF SPLEEN CELLS SENSITIVE TO IRBP

    The spleen cells,from the Liwis rats on the 14th day after immunization with interphotoreceptor retinoid-binding protein(IRBP),were cultured alone or with IRBP for 48 hours.As high as 3times;10 7cells were intraperitoneously transfered to naive rats.The cells stimulated by IRBP in vitro were able to transfer both EAU,EAP and specific immune reactions,contrary to those cultured aloe which transfer neither EAU,EAP nor specific immune reactions.It is highly suggested that EAU is predominantly T cell-mediated and that cells incubation with IRBP prior to transfer is indispensable for induction of EAU in naive recipients.   • Learning Curve of Allogeneic Mouse Model of Peritoneal Heart Transplantation

    Allogeneic mouse model of peritoneal heart transplant is a microscopic surgery on small animal with complex techniques. For a beginner, a learning curve of this surgical technique has to be experienced. The learning curve contains three stages:(1) to be familiar with the local anatomy of either donor or recipient mouse; (2) to be capable of collecting donor heart and well preparing the major peritoneal vessels of recipient; (3) to be skillful in the anastomosis of major vessels. The bottleneck of the learning curve is the valid skill of vascular anastomosis. The stepwise essentials are to "understand, be familiar, be accurate, and be quick" in the learning curve.

  • Simiao Yong'an decoction for the treatment of atherosclerosis in animal experiment: a systematic review

    ObjectiveTo systematically review the interventional effects of Simiao Yong'an decoction on atherosclerosis animal models.MethodsDatabase including CNKI, WanFang Data, VIP, PubMed, The Cochrane Library, and Web of Science were searched to collect animal experiments on atherosclerosis model intervention by Simiao Yong’an decoction from inception to October 2020. Two reviewers independently screened the literature, extracted data, and used the SYRCLE animal experiment bias risk assessment tool to evaluate risk bias of included studies, and then used RevMan 5.4.1 software for meta-analysis.ResultsA total of 14 animal experiments were included. The results of meta-analysis showed that compared with the blank model group, the Simiao Yong’an decoction group could reduce the aortic plaque area (SMD=−2.04, 95%CI −3.35 to −0.74), the ratio of aortic plaque to lumen area (SMD=−1.72, 95%CI −2.48 to −0.97), total cholesterol level (SMD=−0.97, 95 %CI −1.72 to −0.22), triglyceride level (SMD=−1.21, 95%CI −1.82 to −0.60), low-density lipoprotein cholesterol level (SMD=−1.82, 95%CI −3.12 to −1.53), tumor necrosis factor-α level (SMD=−3.36, 95%CI −4.21 to −2.52), monocyte chemotactic factor-1 level (SMD=−2.98, 95%CI −4.60 to −1.35) and C-reactive protein level (SMD=−0.60, 95%CI −1.08 to −0.11); however, in the high-density lipoprotein cholesterol level (SMD=0.66, 95%CI −0.10 to 1.42) and the level of interleukin 1 (SMD=−1.41, 95%CI −4.11 to 1.30), the differences were not statistically significant.ConclusionsThe existing evidence shows that the intervention of Simiao Yong’an decoction in the atherosclerosis model can reduce the aortic plaque area and the ratio of the aortic plaque to the lumen area, reduce total cholesterol, triglycerides, and low-density lipoprotein cholesterol levels, and reduce tumor necrosis factor-α, monocyte chemotactic factor-1, and C-reactive protein levels. Due to limited quality of included studies, more high quality studies are required to verify the above conclusions.

  • IMPLANTATION OF ALLOGENIC OSTEOBLAST COMBINED WITH CALCIUM PHOSPHTA COMPOSITES

    The aim of this experiment was to study the osteogenesis in vivo of allogenic osteoblast combined culture with calcium phosphate composites. The osteoblasts were obtained by enzymatic digestion of periosteum from fibula subcultured to 13 generations, the cells were combined culture with hydroxyapatite and biphasic calcium phosphate. Subseguently, the composite was implanted into rabbits subcutaneously or intramuscularly. The blank material was implanted in the contralateral side as control. Four weeks later, all animals were sacrificed. All the implants were examined by gross observation, histological examination and EDXA. The results showed: 1. obvious ingrowth of connective tissue with very little inflammatory reaction; 2. new bone formation in the composites with deposit of Ca and P on the surface of osteoblast, but none in the blank materials; 3. no significant difference of new bone formation between the different sites of implantation or different materials, but those implanted intramuscularly had lamellae form of new bone while those implanted subcutaneously had only mineralization of extracellular matrix. The conclusion were: 1. the composites are biocompatible with prior osteogenesis property; 2. periosteal-derived allogenic osteoblasts obatined by enzymatic digestion could survive following implantation with bioactivity; 3. rich blood supply might be advantageous to new bone formation and its maturation.

  • EXPERIMENTAL STUDY OF INSTANTANEOUS EXPANSION OF SOFT TISSUE

    Abstract An animal experiment had been conducted for the purpose to find out the possibility of application of instantaneous sustained limited expansion (ISLE). A total of 54 skin defects in 9 pigs were obtained and were divided into two groups. One group of the pigs were subjected to instantaneous sustained limited expansion, the others, as the control group, just received the tension suture. At the same time the blood supply of the skin was measured by laser doppler flowmeter (LDF). It was showed that microcirculatory perfusion of the ISLE group was considerably better than that of the control. Inaddition, the correlation between the blood flow of the skin and the expanding pressure, as well as the pathologic changes of the skin were discussed.

  • Experimental Research of Recombinant Human Growth Hormone on Obstructive Jaundice

    ObjectiveTherapeutical effect of recombinant human growth hormone (rhGH) on obstructive jaundice and internal and external drainage was observed.MethodsNew Zealand white rabbits were randomly divided into groups below: obstructive jaundice internal drainage plus rhGH group, obstructive jaundice internal drainage plus NS group, obstructive jaundice external drainage plus NS group, and obstructive jaundice external drainage plus rhGH group. After the establishment of obstructive jaundice model, rhGH was used in the above groups. Subcutaneous injection of rhGH 0.2 IU/kg was given twice a day. Isovolume NS was used on the control groups. Full set of endotoxin, tumor necrosis factor, sIL2R and nutritional status were estimated before the model establishment, and 14 days after the model established, 14 days after internal and external drainage.ResultsFour days after internal and external drainage, body weight of therapy groups was increased compared with control groups (P<0.05). Seven days and ten days after obstructive jaundice, blood sugar of therapy groups rised compared with control groups (P<0.05). Albuminate, siderophilin and prealbumin of therapy groups were all observed an increase after 14 days after obstructive jaundice, and 14 days after internal and external drainage (P<0.01). Blood total cholesterol, low density lipoprotein and omni bile acid of therapy groups after 14 days of obstructive jaundice were increased apparently (P<0.05). Blood glutamicoxal acetic transaminase, transglutaminase, total bilirubin, blood uria nitrogen, creatinine and uric acid of therapy group after 14 obstructive jaundice days were increased (P<0.05). Ca2+ of therapy groups 14 days after obstructive jaundice, 14 days after internal and external drainage rised as compared with control groups (P<0.05). However, K+,Na+ of therapy groups 14 days after external drainage decreased (P<0.05). An increasing tendency of sIL2R was observed in control groups 14 days after obstructive jaundice(P<0.05) and ET,αTNF,sIL2R of control groups was decreased 14 days after internal and external drainage (P<0.01).ConclusionAfter rhGH is used in obstructive jaundice and internal and external drainage, an improvement of nutritional status and immunological function can be observed.

  • EXPERIMENTAL STUDY ON TENSILE STRENGTH OF CARBONTENDON IMPLANTEDINTO BONE

    A mechanical study on the bones of 29 rabbits following implantation of carbontendon was carried out. The rabbits were divided into seven groups according to the observation time (2,4,6,8,12,20 and 30 weeks after operation). A bundle of artificial tendon composed of 7,000 carbon fibers was passedthrough a tunnel in the tibia, and both ends of the artificial tendon were ligated to the muscle fibers. The mechanical strength and histological structure of the carbonbone junction and their relationship were studied in each group. Carbon fiberwas split and degradated in six to eight weeks after operation. The tensile strength of carbontendon in the soft tissue was decreased from 82±4.6N in the second week to 27±5.31N and6.3±1.81N in the sixth and eighth week respectively. The tensile strength of carbontendon increased from 3.01±1.2N to 6.1±2.01N at the carbon -tendon-bone junction in the bone. The tensile strength of carbon-tendon was unsatisfactory for implantation into bone. The carbon-tendon was split and degradated and the tensile strength was not b enough to cope with the early functional exercises.

  • SEQUENTIAL EXPERIMENTAL STUDY OF DENERVATED NEURO-MUSCULAR JUNCTIO

    Motor endplate is the structure connected between the nerve terminal and muscle fibre and plays a very important role in conducting nerve impulses to the target, therefore, systemical study of the sequential changes of the motor end-plate upon denervation is quite important.Ninety New Zealand rabbits were divided into nine groups from two weeks to nine months after denervation. Acetycholinesterase(AchE) was analyzed quantitatively to study the sequential changes of the motorendplates of tibialis anterior muscle. The results showed that there was no significant reduction of AchE at theend if one month after denervation, whereas a sharp reduction of AchE afterwards. AchE could not be stained after five month denervation.
